JOB DESCRIPTION:

Role & responsibilities:

Experiments:

• Execution of planned experiments/studies using higher-order structure methods.
• Primary structure analysis using amino acid profiling and N-terminal sequencing.
• Amino acid composition analysis using UPLC/HPLC based methods for peptides and proteins
• Secondary structure evaluation using Circular Dichroism (CD) with desalting optimization techniques.
• Amide bond and conformational analysis using FTIR spectroscopy.
• Peptide aggregation and oligomerization studies using SEC and fluorescence-based assays
• Execution of planned experiments/studies using physico-chemical methods.
• Develop advanced LC-MS-based methodologies (ESI, ETD, ECD) for the characterization of protein and peptide products.
• Design and implement integrated analytical strategies by combining chromatography and mass spectrometry techniques. These strategies aim to qualitatively and quantitatively assess product-related variants and impurities. This includes the development of innovative sample preparation methods, with a focus on LC-MS, CEX-MS, and SEC-MS, native MS, HDX as well as the characterization of innovator product batches sourced from various geographical regions to establish a comprehensive quality target profile.
• Lead the development of sophisticated analytical workflows, including multi-attribute methods for post-translational modifications (PTMs), sequence variant analysis, profiling of host cell proteins (HCPs), disulfide bond analysis and shuffling, cell culture metabolite analysis, and glycan profiling.
• Impurity characterization of peptides and comparability assessment with innovator using 2DLC (preferably from Agilent) Qualification of LCMS methods and method transfer according to ICH and regulatory guidelines.
• Fibrillation and amyloid kinetics studies using Thioflavin-T fluorescence assays
• Charge variant/heterogeneity profiling using SCX chromatography
• Hydrophobicity and polarity profiling using RP-HPLC methods.
• Low-level impurity detection (LOQ level) and troubleshooting recovery challenges in peptide analysis.
• Assay, impurities, aggregation, free vs bound drug analysis through different Peptide-related analytical method development

Interpretations:

• Scientific interpretations of processed data (manual/software-driven).
• Stability and degradation pathway assessment of peptides under different stress conditions.
• Regulatory deficiency handling for peptide-based products and analytical justifications
• Establishing quality target profiles according to Q14 and Q2R2/R1
• Scientific interpretations of processed data (manual/software-driven).

Data Generation:

• Sample preparation, data acquisition, data analysis/processing of HPLC/UPLC/LCMS/HRMS/CE based experiments/studies. Preferably on Thermo/Waters/Sciex platforms.
• Expertise on techniques such as CD/FTIR/SEC-FFF-MALS/DSC/DLS as well as different separation technology based on HPLC and UPLC.
