During my M1 internship, I focused on assessing the potential role of telomeric repeats in regulating epigenome homeostasis in the model plant Arabidopsis thaliana. Our study aimed to understand how telomeric repeat binding proteins (TRBs) influence the chromatin environment at interstitial telomeric repeats (ITRs), regions similar to telomeres but located within the genome during de-etiolation, the transition of a plant from a dark-grown state to a light-grown state. Through phenotyping, gene expression analysis (RT-qPCR), and pigment analysis experiments on wild-type and mutant A. thaliana lines, including those lacking TRB proteins, I investigated the impact of TRB gene mutations on plant phenotype and epigenetic profiles. Our findings highlighted TRBs' potential role in regulating the morphological and metabolic changes during the de-etiolation process. This research not only expanded our understanding of chromatin regulation in plants but also proposed implications for evolutionary biology.
