Oct 2012 - Sep 2013
1 year
Pirbright, UK
Working on the Rapidia-FIELD project on the use of LAMP (Loop Mediated Isothermal Amplification) to identify livestock animals with Foot and Mouth Disease or Swine Vesicular Disease. More specifically, tested and optimized the published versions of the LAMP assay for the two viruses, adapted the assay to be run on the portable Genie II machine for use on the field, and set up a multiplex assay to test for both FMDV and SVDV via LAMP and Lateral Flow Device using fluorescence labelled primers. On the side, compared and analyzed new sequences found in this laboratory for SVDV, performed a sliding window analysis (SWAN) for variability and confirmed that the primers being used for the SVDV LAMP were still the best ones according to the new sequences. Key points: – High containment laboratory rules and SOPs – Multiplexing Foot-and-Mouth Disease and Swine Vesicular Disease – Primer design and alignment – RNA extraction from supernatant via purification kits – RT-PCR and RT-LAMP – Dry down assay for RT-PCR – Gel electrophoresis – Lateral Flow Device – Sensitivity tests – RNA quantitation via NanoDrop – Assay Development and Optimization Data presented at EUFMD Croatia 2014

Four weeks of apprenticeship and hands on experience in four different laboratory sectors: cell cultures, GMO testing, bacterial virology, and DNA sequencing. The internship was very educational, as I learned both the theory behind the use of certain techniques, and how to use the techniques themselves, along with the appropriate machines involved in DNA sequencing and PCR reactions
