– Stress is one of the key role player leading to arousal, valence and engagement of the person. In the project we are measuring the stress within the iGEM teams during the important final deadline of their project, the wiki freeze. – For that purpose, we will record the level of stress using a mobile application that measures heart rate with the smartphone camera, without the need for external pulsometers. – This app asks the user to press the smartphone camera lens gently with their finger and, after some seconds, a reading is shown. Then integration of this data with stress hormones level and their engagement, commitment, integration in the team. – In this way we are using a holistic approach to understand team behaviours and the most smart way to tackle the stress.

– iGEM Paris Bettencourt Foundation 2018: Antibiotic overuse in livestock industry is one of the major drivers to the antibiotic resistance evolution; motivating calls to reduce, replace, and re-think the antibiotic usage in animals. – Antimicrobial peptides (AMPs) are a promising alternative to conventional antibiotics. – Recently, a class of chemically-synthesized, star-shaped AMPs has been shown to exhibit broad-spectrum antimicrobial activity while maintaining biocompatibility with mammalian cells. – In this project, we combinatorially fused a set of known AMPs to structurally diverse, self-assembling protein cores to produce star-shaped complexes. – Over 200 fusions were designed and expressed in a cell-free system, then screened for activity, biocompatibility, and membrane selectivity. – In addition, we selected 4 AMPs for rational mutagenesis (~12000 variants), and a subset of fusions for molecular dynamic modeling, to identify features of surface charge and star geometry that impact AMP performance. – Overall, we aim to create a novel class of selective, non-toxic AMPs which are biologically-produced.

– The group developed an E. coli strain that allows the growth rate to be switched on and off externally by adjusting the availability of RNA polymerase, a major component of the gene expression machinery. – It was shown that by arresting the expression of RNA polymerase resources are shuttled away from protein synthesis (growth) to the production of a metabolite of interest (glycerol). – This proof-of-principle has been extended from the microliter to the liter scale, in collaboration with a biotechnology company, and we have acquired metabolomics, transcriptomics, and proteomics datasets to characterize the physiology of both growing and growth-arrested, glycerol-producing cells. – My project was focused on the integration of these datasets using a combination of statistical techniques and a genome-scale flux balance models of E. coli metabolism, in order to study the reorientation of fluxes from growth to glycerol production on the molecular level and the possible occurrence of stress responses. – The conclusions drawn from this study lead to further validation experiments with the growth switch. Moreoever, the improvement of some aspects of the system, such as its genetic stability or the induction mechanism, were also explored during the internship time.
